Her paternal grandparents were Thomas le Despenser and Constance of York, linking her to significant English aristocratic lineages.

Through her marriage to Richard Neville, she became the mother of two daughters who would play pivotal roles in English history. Her elder daughter, Isabel Neville, became the Duchess of Clarence through marriage to George Plantagenet, while her younger daughter, Anne Neville, ascended to the throne as the Queen of England by marrying King Richard III.
