Isabel Marshal was born on 9 October 1200 at Pembroke Castle, Wales, as the seventh child of William Marshal, 1st Earl of Pembroke, and Isabel de Clare. She was part of the Marshal family and had nine siblings, including brothers who held the title of Earl of Pembroke. Her sisters married into noble families, such as the Earls of Norfolk, Surrey, and Derby.

On her 17th birthday, Isabel married Gilbert de Clare, 4th Earl of Hertford and 5th Earl of Gloucester, at Tewkesbury Abbey. They had five children together. After Gilbert's death in 1230, Isabel married Richard, 1st Earl of Cornwall, in 1231, and they had four children, although three died in infancy.

Isabel died at the age of 39 on 17 January 1240 at Berkhamsted Castle due to liver failure contracted during childbirth. She was buried at Beaulieu Abbey, and her heart was sent to Tewkesbury Abbey.
